If I were to purchase a Trend Micro or MacAfee one year subscription could it be used on my desktop and laptop? Or would I have to purchase two separate subscriptions?
Also, which one is better to use with Vista?

For a single-user subscription, it is only applicable to ONE particular computer system: desktop OR laptop, not both. For volume-license it could be used on a NUMBER of computer systems. Thus, if you purchase a single-user subscription, then it is only for the first computer system it was originally installed.

For Vista, try AVG 8.0, you could even try it for free http://free.avg.com
