To uninstall programs go to start—control panel—add or remove programs.

if the files you want to remove is some programs than you have to delete via this procedure:
control panel –> add/remove program (windows xp) / program than uninstall (in vista) –> than uninstall that program and than you get rid of that files.
if want to remove a regular files that are of your personal and somehow operating system is denying of removeal than start the computer in safe mode (press F8 as you press the computer power button than chose safe mode)
after booting through safe mode now you can be able to delete that files.
than after deleteing you can continue in normal mode after restating the pc.
and still you aren't able to delete that files that may means that the files are placed in unreadble normally bad sector assumed area of the disk drive. than you can do any thing.
MOST OF THE TIMES OPERATING SYSTEM DENIED OF DELETION OF FILES BECAUSE THEY ARE BEEN NEEDED BY OPERATING SYSTEM THAT MEANS THEY ARE SYSTEM FILE AND IF THEY ARE MADE BY YOU THAN YOU CAN DELETE THEM THROUGH SAFE MODE, SAFELY
