Removing a trojan?

Yes i know run anti virus programs well they arent detecting it. I have tried all of the anti virus programs. I know the exact file where the trojan is, but i need this file. I try deleting the file and redownloading it (from different sources) and the trojan comes back! Any ideas on how to get rid of it? Also how do the professionals remove trojans/viruses from a computer?
